Canonically, Thor can’t detect Loki’s illusions. He fell for them in both the previous movies, at times when it was a lot more important than it is here.
Maybe he had the habit of just randomly saying this to Loki sometimes in case Loki happened to be casting an illusion right then?

To me, for that reason, this scene was actually one of those very subtle moments that demonstrated that Thor is not entirely hopeless when it comes to showing his brother affection and decency.
